The most cost effective way is to do the trip at the start or end of the holiday and combine it within your transatlantic tickets on the same airline. Not only will the fare be much cheaper but you won't be paying for 2 lots of accommodation while you're in New York.
If you really want to do it in the middle of the holiday, I'd advise booking in the UK before you go. Expect to pay anything between about £120 & £200 each for a direct flight.
We flew with Delta to Orlando via Atlanta a couple of Christmases ago and on the way back broke our journey to spend 4 days over Christmas in New York. We did not pay any more to do this - we simply flew from Orlando to JFK, broke our return journey for 4 days, and then flew home from there.
My daughter flew in to New York on the way over and got a cheap internal flight down to Orlando, so maybe you could pick up a flight on one of the budget airlines to and from New York if you wanted to break up your stay in Orlando.
However, as KG says, it is more cost effective to add it on to either the beginning of the end of your trip as then you won't be paying extra for these flights, and you won't be paying twice for accommodation.

We often visit New York and normally do it either on our way to the villa or on the way back. As others have said, it is the cheapest way to do it as you can have the fare as part of your trans-atlantic ticket and just break your journey and have a stopover.
Sometimes it can be cheaper to book a return flight from the UK to New York or even go to New York and come back from Orlando direct to the UK. These can be good if there are special deals. You can then get a cheap internal flight on an airline like Jet Blue between New York and Orlando. We did this last year and got an internal flight for just $97 on Jet Blue.
We have also gone up for a long weekend in the middle of our stay in the villa to see special concerts. Lots of cheap flights but we have always ended up booking Jet Blue as they have worked out the best on price. Just because they are cheap don't think that they are basic. Great service, in flight snack and drink and seat back TVs showing live TV.
New York is one of out all time favourite places to visit.
